In my organization we have 6 persons in ESI slab as per new rule (i.e. Gross up to 15000 from May 2010) and we don't have ESI no for our company. Total employee strength comes up to 30. Should we deduct esi for these 6 employees? And what is the procedure to get ESI No. for the organization.
These 6 emploees are not direct employee they under direct contract employee

Minimum Employees for ESIC is 20, whom falls under the ESIC ceiling/salary range. If you don't have that no., then you need not register. This applies to full time employees and not contract employment.
minimum employees is 10 or more, if your establishment is undergone with the aid of power(manufacturing process), otherwise its 20 , and in your case company has to ensure that whether contractor is facilitating ESI to those employees..
